I nominate Sean Connery as James Bond. All other Bonds are inevitably compared to Mr. Connery's. Perhaps it was just Sean Connery's take on the character, but he gave us the definitive version of who James Bond is. There have been five James Bonds since Connery, (not counting the original Woody Allen Casino Royale as a real Bond movie) but when I think 007, the only actor I can associate with the character really is Connery.
 
Edward Norton as Derek Vinyard- Orton owned the role as Vinyard in American History X, it's extremely hard to imagine anyone else in that role and them being able to even get the slightest bit of sympathy that he got for his character as his brother get dragged into the world he left.

This is my choice also. Norton didn't just own that role, he made it his bitch.

I thought about putting Jay from the Kevin Smith movies as my first choice. Most notably from Clerks 2. He may not be the best actor, but I don't think anyone could take his place. They guy is almost exactly the same person off camera as he is on camera.

That's not really acting then. That's just reading lines. Which is pretty much what The Rock does, and he's never owned any role. Any true actor could replace Jason Mewes. In fact a proper actor could do a better Jason Mewes than Jason Mews could.
 
Javier Bardem - Anton Chigurh in "No Country for Old Men"

I didn't care for the movie. I love the Coen Brothers, but to me, it was very slow-paced, and the ending was horrible. However, Bardem as Anton was just an awesome role. The gas station scene with the quarter stands out to me more than anything, but the way he carried himself in that role was the reason why I watched the entire movie, when I would have otherwise left the theater.

Bardem played Anton as a complete psychopath. He was incredibly violent, and that hair ruled. He didn't speak that often in the movie, but when he did, he spoke with true evil in his voice.
