Santino made up an origin story for the Cobra today on WWE Inbox, but I could've sworn Jon Lovitz had taught it to him on Raw.
After looking it up, it seems like Lovitz taught him a submission maneuver called the "Fast Freak Fiber Twitch Jugular Pinch" where Santino would pinch someone's jugular. He told him he had to be "faster than a cobra" to hit it correctly. Santino wouldn't hit it like a strike as he does today, but I think it's clear that this was the start of the Cobra. http://www.hulu.com/watch/152180
